This sequence change affects a splice site in intron 7 of the MTHFR gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Studies have shown that disruption of this splice site results in skipping of exon 8 and introduces a premature termination codon (PMID: 25736335). The resulting mRNA is expected to undergo nonsense-mediated decay. ClinVar contains an entry for this variant (Variation ID: 187891). This variant is also known as c.1179-2delA. Disruption of this splice site has been observed in individuals with severe MTHFR deficiency (PMID: 25736335). This variant is present in population databases (rs780014899, gnomAD 0.002%). -
