The p.G285R variant (also known as c.853G>A), located in coding exon 15 of the TNNT2 gene, results from a G to A substitution at nucleotide position 853. The glycine at codon 285 is replaced by arginine, an amino acid with dissimilar properties. This alteration has been reported in a hypertrophic cardiomyopathy (HCM) cohort; however, clinical details were limited (Kim HY et al. J Clin Med, 2020 Jun;9:[ePub ahead of print]). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
