Variant summary: TNNT2 c.224T>G (p.Val75Gly) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251036 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.224T>G in individuals affected with Cardiomyopathy and no experimental evidence demonstrating its impact on protein function have been reported. Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -

This missense variant replaces valine with glycine at codon 75 of the TNNT2 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with dilated cardiomyopathy (PMID: 34935411). This variant has been identified in 2/251036 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The p.V75G variant (also known as c.224T>G), located in coding exon 7 of the TNNT2 gene, results from a T to G substitution at nucleotide position 224. The valine at codon 75 is replaced by glycine, an amino acid with dissimilar properties. This alteration has been reported in a population-based longitudinal study in an individual without overt cardiomyopathy, and in an individual from a pediatric dilated cardiomyopathy cohort (Bick AG et al. Am J Hum Genet, 2012 Sep;91:513-9; Khan RS et al. J Am Heart Assoc. 2022 Jan;11(1):e022854). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
