Their diverse functions include regulating neurotransmitter release, heart rate, insulin secretion, neuronal excitability, epithelial electrolyte transport, smooth muscle contraction, and cell volume. This gene encodes a member of the potassium channel, voltage-gated, subfamily H. This member is a pore-forming (alpha) subunit of a voltage-gated non-inactivating delayed rectifier potassium channel. It is activated at the onset of myoblast differentiation. The gene is highly expressed in brain and in myoblasts. Overexpression of the gene may confer a growth advantage to cancer cells and favor tumor cell proliferation. Alternative splicing of this gene results in two transcript variants encoding distinct isoforms. [provided by RefSeq, Jul 2008]

The variant allele was found at a frequency of 2.2e-05 in 1613768 control chromosomes (i.e. 36 individuals) in the gnomAD database (v4.0), suggesting it is not likely to be associated with a highly penetrant autosomal dominant condition. c.2942C>G has been reported in the literature in an individual with autism, without strong evidence for causality (Wilfert_2021). This report does not provide unequivocal conclusions about association of the variant with KCNH1-Related Disorders. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 34312540). One submitter has cited a clinical-significance assessment for this variant to ClinVar after 2014 and has classified the variant as likely benign.
