This variant has not been reported in the literature in individuals affected with NEK2-related conditions. This sequence change replaces lysine, which is basic and polar, with arginine, which is basic and polar, at codon 409 of the NEK2 protein (p.Lys409Arg). This variant is present in population databases (rs576980203, gnomAD 0.007%). ClinVar contains an entry for this variant (Variation ID: 1945208). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
