A functional study has reported that transgenic mice homozygous for this variant show mild cardiac hypertrophy and an increased susceptibility to arrhythmia before the onset of hypertrophy (PMID: 30835254). This variant has been reported in an individual affected with hypertrophic cardiomyopathy (PMID: 30835254). This variant has been identified in 7/249216 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The p.P1124L variant (also known as c.3371C>T), located in coding exon 28 of the RYR2 gene, results from a C to T substitution at nucleotide position 3371. The proline at codon 1124 is replaced by leucine, an amino acid with similar properties. This alteration has been reported in a subject with hypertrophic cardiomyopathy (HCM) and may have an impact on protein function (Alvarado FJ et al. JCI Insight, 2019 03;5:). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
