The arginine at codon 3510 is replaced by cysteine, an amino acid with highly dissimilar properties. This alteration has been detected in a hypertrophic cardiomyopathy cohort, but clinical details were limited (Mademont-Soler I et al. PLoS ONE. 2017;12:e0181465). This variant has also been reported as a secondary cardiac variant in exome cohorts (Ng D et al. Circ Cardiovasc Genet. 2013;6:337-46; Landstrom AP et al. Circ Arrhythm Electrophysiol. 2017;10:e004742). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
