The variant received 0 classification points (ACGS-UK Somatic Oncogenicity v2025).

The NM_001035.3(RYR2):c.13291G>A (p.Glu4431Lys) variant causes a missense change involving the alteration of a non-conserved nucleotide. The gene RYR2 is a tumor suppressor gene (CancerMine: 2 TSG citations). The variant allele was found at a cumulative frequency of 0.000558 (AC=889) in the gnomAD database across 1,591,854 control chromosomes (no homozygotes observed). The grpmax filtering allele frequency (95% CI) is 0.000704. Splicing prediction tools (SpliceAI) predict no significant impact on normal splicing. Variant has been reported in ClinVar as Benign/Likely Benign (no review stars). Other variants at the same amino acid position have been reported in ClinVar (not pathogenic): p.E4431= (synonymous): Benign/Likely_benign (ClinVar VariationId 496090, 2 stars) The variant has been observed in cBioPortal in 1 sample across 1 study and 1 cancer type; somatic enrichment level: NONE (Observed in at most one deduplicated cBioPortal case.). This exact variant is curated in the UniProt human variants database as Uncertain Significance.

Genes affected
RYR2 (HGNC:10484): (ryanodine receptor 2) This gene encodes a ryanodine receptor found in cardiac muscle sarcoplasmic reticulum. The encoded protein is one of the components of a calcium channel, composed of a tetramer of the ryanodine receptor proteins and a tetramer of FK506 binding protein 1B proteins, that supplies calcium to cardiac muscle. Mutations in this gene are associated with stress-induced polymorphic ventricular tachycardia and arrhythmogenic right ventricular dysplasia. [provided by RefSeq, Jul 2008]
