P3H1 (HGNC:19316): (prolyl 3-hydroxylase 1) This gene encodes an enzyme that is a member of the collagen prolyl hydroxylase family. These enzymes are localized to the endoplasmic reticulum and their activity is required for proper collagen synthesis and assembly. Mutations in this gene are associated with osteogenesis imperfecta type VIII. Three alternatively spliced transcript variants encoding different isoforms have been described. This sequence change falls in intron 14 of the P3H1 gene. It does not directly change the encoded amino acid sequence of the P3H1 protein. RNA analysis indicates that this variant induces altered splicing and likely disrupts the C-terminus of the protein. This variant is present in population databases (rs137853890, gnomAD 0.0008%). This variant has been observed in individuals with autosomal recessive osteogenesis imperfecta (PMID: 3545499, 22615817, 27864101). ClinVar contains an entry for this variant (Variation ID: 1259). Studies have shown that this variant results in strengthens a cryptic splice site in intron 14 and the retention of a 19 bp repeat and introduces a new termination codon (PMID: 19088120). However the mRNA is not expected to undergo nonsense-mediated decay. This variant disrupts the C-terminus of the P3H1 protein. Other variant(s) that disrupt this region (p.Q722*, p.Glu719Argfs*11) have been observed in individuals with P3H1-related conditions (PMID: 22615817, 27864101). This suggests that this may be a clinically significant region of the protein. For these reasons, this variant has been classified as Pathogenic. -
