This sequence change replaces arginine, which is basic and polar, with glycine, which is neutral and non-polar, at codon 437 of the MUTYH protein (p.Arg437Gly). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Hereditary cancer-predisposing syndrome Uncertain:1
The p.R437G variant (also known as c.1309C>G), located in coding exon 13 of the MUTYH gene, results from a C to G substitution at nucleotide position 1309. The arginine at codon 437 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
