Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 248926 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.53G>A has been reported in the Euroglycan network in association with CDG (Haeuptle_2009, Morava_2016) and in the literature in one compound heterozygous individual affected with Congenital Disorder Of Glycosylation (Vleugels_2011). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as VUS (n=2) and as likely pathogenic (n=1).
