The homotetramer enzyme catalyzes the initial step of the mitochondrial fatty acid beta-oxidation pathway. Defects in this gene cause medium-chain acyl-CoA dehydrogenase deficiency, a disease characterized by hepatic dysfunction, fasting hypoglycemia, and encephalopathy, which can result in infantile death. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 7.2e-05 in 251166 control chromosomes. This frequency is not significantly higher than expected for a pathogenic variant in ACADM causing Medium Chain Acyl-CoA Dehydrogenase Deficiency (7.2e-05 vs 0.0054), allowing no conclusion about variant significance. c.250C>T has been reported in the literature in multiple compound heterozygous individuals affected with Medium Chain Acyl-CoA Dehydrogenase Deficiency (Clayton_1998, Waddell_2006, Hsu_2008, Kennedy_2010, Smith_2010, Tangeraas_2020). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
