Hermansky-Pudlak syndrome 6 Pathogenic:1
- -
not provided Pathogenic:1
This variant disrupts a region of the HPS6 protein in which other variant(s) (p.Gln680*) have been determined to be pathogenic (PMID: 27225848). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. ClinVar contains an entry for this variant (Variation ID: 435465). This variant has not been reported in the literature in individuals affected with HPS6-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Cys571*) in the HPS6 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 205 amino acid(s) of the HPS6 protein. For these reasons, this variant has been classified as Pathogenic. -
Hermansky-Pudlak syndrome Pathogenic:1
Variant summary: HPS6 c.1711_1712insAG (p.Cys571X), located in exon 1 of the single exon gene, results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been reported in association with Hermansky-Pudlak Syndrome in the HGMD database. The variant was absent in 248294 control chromosomes. c.1711_1712insAG has been reported in the literature among clinically significant variants in individuals affected with Hermansky-Pudlak Syndrome (example, Karim_2022). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as likely pathogenic. -
