Genes affected
RBM20 (HGNC:27424): (RNA binding motif protein 20) This gene encodes a protein that binds RNA and regulates splicing. Mutations in this gene have been associated with familial dilated cardiomyopathy. [provided by RefSeq, Apr 2014]

Uncertain significance, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineSep 30, 2014The Lys420Arg variant in RBM20 gene has not been previously reported in individu als with cardiomyopathy or in large population studies. Computational prediction tools and conservation analysis suggest that the Lys420Arg variant may not impa ct the protein, though this information is not predictive enough to rule out pat hogenicity. In summary, the clinical significance of the Lys420Arg variant is un certain. -
Cardiovascular phenotype Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingAmbry GeneticsAug 26, 2020The p.K420R variant (also known as c.1259A>G), located in coding exon 2 of the RBM20 gene, results from an A to G substitution at nucleotide position 1259. The lysine at codon 420 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
