The c.1003A>C (p.S335R) alteration is located in exon 10 (coding exon 10) of the NEBL gene. This alteration results from a A to C substitution at nucleotide position 1003, causing the serine (S) at amino acid position 335 to be replaced by an arginine (R). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces serine with arginine at codon 335 of the NEBL protein (p.Ser335Arg). The serine residue is highly conserved and there is a moderate physicochemical difference between serine and arginine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has not been reported in the literature in individuals with NEBL-related disease.
