Variant classified as Uncertain Significance - Favor Benign. The p.Met1014Val va riant in MYO3A has not been previously reported in individuals with hearing loss or in large population studies. The methionine (Met) at position 1014 is not co nserved in mammals or evolutionary distant species, and >10 mammals carry a vali ne (Val), suggesting that this change at this position may be tolerated. However , nearby amino acids are not highly conserved either. Computational tools predic t the creation of a possible cryptic splice site; however, this information is n ot predictive enough to determine pathogenicity.
