This missense variant replaces arginine with glutamine at codon 57 of the RET protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with RET-related disorders in the literature. This variant has been identified in 6/250982 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 57 of the RET protein (p.Arg57Gln). This variant is present in population databases (rs779915615, gnomAD 0.006%). This variant has been reported in a fetus affected with multicystic dysplastic kidney, as well as the unaffected father (PMID: 21490379). ClinVar contains an entry for this variant (Variation ID: 477323). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign".
