This sequence change replaces aspartic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 624 of the RET protein (p.Asp624Gly). The p.D624G variant (also known as c.1871A>G), located in coding exon 10 of the RET gene, results from an A to G substitution at nucleotide position 1871. The aspartic acid at codon 624 is replaced by glycine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
