To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with medullary thyroid cancer and pheochromocytoma (PMID: 28946813). This variant has been identified in 1/250868 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Different missense variants occurring at the same codon, p.Lys666Asn and p.Lys666Glu, are well-documented pathogenic mutations (ClinVar Variation ID: 230926, 24932, 24931), indicating that lysine at this position is important for RET protein function. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |
