This missense variant replaces asparagine with serine at codon 783 of the RET protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with medullary thyroid cancer (PMID: 28946813) and an individual affected with isolated primary hyperparathyroidism (PMID: 33340421). This variant has been identified in 1/251344 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 783 of the RET protein (p.Asn783Ser). The p.N783S variant (also known as c.2348A>G), located in coding exon 13 of the RET gene, results from an A to G substitution at nucleotide position 2348. The asparagine at codon 783 is replaced by serine, an amino acid with highly similar properties. This alteration has been identified in multiple Chinese patients diagnosed with Hirschsprung disease as well as one French patient diagnosed with medullary thyroid cancer at age 57 (So MT et al. PLoS ONE. 2011 Dec;6:e28986; Tang CS et al. Gastroenterology. 2018 12;155:1908-1922.e5; Lebeault M et al. Thyroid. 2017 12;27:1511-1522). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
