Genes affected
PCDH15 (HGNC:14674): (protocadherin related 15) This gene is a member of the cadherin superfamily. Family members encode integral membrane proteins that mediate calcium-dependent cell-cell adhesion. It plays an essential role in maintenance of normal retinal and cochlear function. Mutations in this gene result in hearing loss and Usher Syndrome Type IF (USH1F). Extensive alternative splicing resulting in multiple isoforms has been observed in the mouse ortholog. Similar alternatively spliced transcripts are inferred to occur in human, and additional variants are likely to occur. [provided by RefSeq, Dec 2008]

Uncertain significance, criteria provided, single submitterclinical testingAthena DiagnosticsJan 05, 2023Available data are insufficient to determine the clinical significance of the variant at this time. The frequency of this variant in the general population is uninformative in assessment of its pathogenicity (http://gnomad.broadinstitute.org). Computational tools yielded predictions that this variant may interfere with normal RNA splicing. -

Uncertain significance, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineOct 16, 2013Variant classified as Uncertain Significance - Favor Benign. The 3807-6T>G varia nt in PCDH15 has not been reported in the literature and data from large populat ion studies is insufficient to assess the frequency of this variant. This varian t is located in the 3' splice region but not in the invariant -1/-2 splice site positions of the splice site consensus sequence. Computational tools do not sugg est an impact to splicing, though this information is not predictive enough to r ule out pathogenicity. In summary, the clinical significance of this variant can not be determined with certainty; however, based upon the computational data, we lean towards a more likely benign role. -
