Z-lines in striated muscle are a sign of this organization, representing the ends of actin thin filaments, titin, nebulin or nebulette and accessory proteins required for structure and function. This gene encodes a protein which interacts with nebulin in skeletal muscle or nebulette in cardiac muscle and alpha-actinin. In addition, this gene product can interact with a protein with the I-band indicating it has a regulatory as well as structural function. Alternative splicing results in multiple transcript variants. p.Ile754Leu in exon 12 of MYPN: This variant is not expected to have clinical si gnificance because it has been identified in 0.30% (57/18866) of East Asian chro mosomes by the genome Aggregation Database (gnomAD, http://gnomad.broadinstitute .org/; dbSNP rs201245117). This variant also has a lack of conservation across s pecies, including mammals. Of note, brush tailed-rat, pika, and shrew have a leu cine (Leu) at this position despite high nearby amino acid conservation.
