CDH23 (HGNC:13733): (cadherin related 23) This gene is a member of the cadherin superfamily, whose genes encode calcium dependent cell-cell adhesion glycoproteins. The encoded protein is thought to be involved in stereocilia organization and hair bundle formation. The gene is located in a region containing the human deafness loci DFNB12 and USH1D. Usher syndrome 1D and nonsyndromic autosomal recessive deafness DFNB12 are caused by allelic mutations of this cadherin-like gene. Upregulation of this gene may also be associated with breast cancer. The Gly2539Arg variant in CDH23 has not been reported in the literature nor prev iously identified by our laboratory. This variant has been identified in 1/8346 (0.01%) European American chromosomes from a broad, though clinically unspecifie d population (NHLBI Exome Sequencing Project; http://evs.gs.washington.edu/EVS). Although this variant has been seen in the general population, its frequency is not high enough to rule out a pathogenic role. Computational analyses (biochemi cal amino acid properties, conservation, AlignGVGD, PolyPhen2, and SIFT) suggest that the Gly2539Arg variant may impact the protein, though this information is not predictive enough to determine pathogenicity. In summary, additional data is needed to determine the clinical significance of this variant. -

This sequence change replaces glycine with arginine at codon 2539 of the CDH23 protein (p.Gly2539Arg). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and arginine. This variant is present in population databases (rs373649718, ExAC 0.009%). This variant has not been reported in the literature in individuals with CDH23-related conditions. ClinVar contains an entry for this variant (Variation ID: 46035). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Not Available"; Align-GVGD: "Class C15").
