The p.S170T variant (also known as c.509G>C), located in coding exon 6 of the PTEN gene, results from a G to C substitution at nucleotide position 509. The serine at codon 170 is replaced by threonine, an amino acid with similar properties. This alteration was detected in a pediatric patient with macrocephaly, suspected austim-spectrum disorder, mild global developmental delay, and megalencephaly, polymicrogyria, and periventricular white matter signal abnormalities on MRI (Yeung KS et al. Mol Autism, 2017 Dec;8:66). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
