This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 29 of the PDE6C protein (p.Arg29Trp). This variant is present in population databases (rs121918537, gnomAD 0.008%). This missense change has been observed in individual(s) with PDE6C-related conditions (PMID: 19615668, 21127010, 30080950). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 8763). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects PDE6C function (PMID: 21127010). For these reasons, this variant has been classified as Pathogenic. -
