This variant has been observed in homozygosity in several individuals affected with achromatopsia or cone-rod dystrophy (PMID: 30080950, Invitae). ClinVar contains an entry for this variant (Variation ID: 424958). This variant is not present in population databases (ExAC no frequency). This sequence change replaces isoleucine with asparagine at codon 714 of the PDE6C protein (p.Ile714Asn). The isoleucine residue is highly conserved and there is a large physicochemical difference between isoleucine and asparagine. -
