Genes affected
PDE6C (HGNC:8787): (phosphodiesterase 6C) This gene encodes the alpha-prime subunit of cone phosphodiesterase, which is composed of a homodimer of two alpha-prime subunits and 3 smaller proteins of 11, 13, and 15 kDa. Mutations in this gene are associated with cone dystrophy type 4 (COD4). [provided by RefSeq, Mar 2010]

Likely pathogenic, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Aug 05, 2019 | In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has been observed in homozygosity in several individuals affected with achromatopsia or cone-rod dystrophy (PMID: 30080950, Invitae). ClinVar contains an entry for this variant (Variation ID: 424958). This variant is not present in population databases (ExAC no frequency). This sequence change replaces isoleucine with asparagine at codon 714 of the PDE6C protein (p.Ile714Asn). The isoleucine residue is highly conserved and there is a large physicochemical difference between isoleucine and asparagine. - |
