However, these predictions have yet to be confirmed by functional studies. The variant was absent in 281798 control chromosomes (gnomAD). A similar variant c.903T>G [p.Gly301Gly] with similar predicted splicing effects was found in 1/7325 healthy European American women over the age of 70 with no personal history of cancer (FLOSSIES database). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.903T>C in individuals affected with Ataxia-Telangiectasia or cancer, and no experimental evidence demonstrating its impact on protein function have been reported. Based on the evidence outlined above, the variant was classified a VUS. - |
