In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with ATM-related conditions. ClinVar contains an entry for this variant (Variation ID: 482549). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. -

The c.7499_7502delTCAAinsCCAG variant, located in coding exon 49 of the ATM gene, results from an in-frame deletion of TCAA and insertion of CCAG at nucleotide positions 7499 to 7502. This results in the deletion of a valine and asparagine residue and the insertion of an alanine and serine residue from codons 2500 to 2501. This amino acid position is highly conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
