This nucleotide position is highly conserved in available vertebrate species. This alteration was not observed in 7051 unselected female breast cancer patients and was observed with an allele frequency of 0.00009 female controls of Japanese ancestry (Momozawa Y et al. Nat Commun, 2018 10;9:4083). In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing; however, this alteration leads to the use of an in frame alternate splice acceptor site (Ambry internal data). The clinical relevance of this splicing event is unknown at this time. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
