The serine at codon 73 is replaced by glycine, an amino acid with similar properties. This variant was detected in 2/314 Chinese pheochromocytoma or paraganglioma (PPGL) patients (Ma X et al. Front Endocrinol (Lausanne), 2020 Dec;11:574662). This variant was also identified in 1/190 unrelated Chinese patients under the age of 45 who presented with renal tumors (Wu J et al. Cancer, 2019 04;125:1060-1069). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
