SDHD (HGNC:10683): (succinate dehydrogenase complex subunit D) This gene encodes a member of complex II of the respiratory chain, which is responsible for the oxidation of succinate. The encoded protein is one of two integral membrane proteins anchoring the complex to the matrix side of the mitochondrial inner membrane. Mutations in this gene are associated with the formation of tumors, including hereditary paraganglioma. Transmission of disease occurs almost exclusively through the paternal allele, suggesting that this locus may be maternally imprinted. There are pseudogenes for this gene on chromosomes 1, 2, 3, 7, and 18. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Feb 2013]

This sequence change deletes 1 nucleotide from exon 4 of the SDHD mRNA (c.394delT), causing a frameshift at codon 132. This creates a premature translational stop signal in the last exon of the SDHD mRNA (p.Ser132Glnfs*3). While this is not anticipated to result in nonsense mediated decay, it is expected to result in a truncated SDHD protein. While this particular variant has not been reported in the literature, a missense substitution downstream of this variant (p.Leu139Pro) has been determined to be pathogenic (PMID: 21348866), suggesting that the leucine 139 residue is critical for SDHD protein function. In summary, this sequence change results in a frameshift and truncation in the last exon of the SDHD gene, and is located upstream of a residue which is required for SDHD protein function. For these reasons, this variant has been classified as Likely Pathogenic. -

The c.394delT pathogenic mutation, located in coding exon 4 of the SDHD gene, results from a deletion of one nucleotide at nucleotide position 394, causing a translational frameshift with a predicted alternate stop codon (p.S132Qfs*3). This alteration occurs at the 3' terminus of theSDHD gene, is not expected to trigger nonsense-mediated mRNA decay, and impacts the last 17.5% of the protein. However, premature stop codons are typically deleterious in nature and the impacted region is critical for protein function and a significant portion of the protein is affected (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD).
