This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 916 of the LRP4 protein (p.Arg916Cys). This variant is present in population databases (rs147259655, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with LRP4-related conditions. ClinVar contains an entry for this variant (Variation ID: 535801). Variant summary: LRP4 c.2746C>T (p.Arg916Cys) results in a non-conservative amino acid change located in the Low-density lipoprotein-receptor YWTD domain (IPR000033) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.8e-05 in 251468 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2746C>T in individuals affected with LRP4-Related Disorders and no experimental evidence demonstrating its impact on protein function have been reported.
