This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 853 of the MYBPC3 protein (p.Gly853Ser). This variant is present in population databases (rs397515976, gnomAD 0.01%). This missense change has been observed in individual(s) with hypertrophic cardiomyopathy (PMID: 25611685, 27532257, 37652022). ClinVar contains an entry for this variant (Variation ID: 42640). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
This missense variant replaces glycine with serine at codon 853 of the MYBPC3 protein. The Gly853Ser variant in MYBPC3 has not been reported in the literature nor prev iously identified by our laboratory. This variant has not been identified in a v ery large and broad population by the NHLBI Exome Sequencing Project (http://evs .gs.washington.edu/EVS). This low frequency is consistent with a disease causing role but insufficient to establish this with confidence. Glycine (Gly) at posit ion 853 is highly conserved in evolution, suggesting that a change would not be tolerated. Other computational analyses (biochemical amino acid properties, Alig nGVGD, PolyPhen2, and SIFT) do not provide strong support for or against an impa ct to the protein. Additional information is needed to fully assess the clinical significance of the Gly853Ser variant. -
