The c.1010G>A (p.G337E) alteration is located in exon 10 (coding exon 9) of the BSCL2 gene. This alteration results from a G to A substitution at nucleotide position 1010, causing the glycine (G) at amino acid position 337 to be replaced by a glutamic acid (E). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Charcot-Marie-Tooth disease type 2 Uncertain:1
This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 337 of the BSCL2 protein (p.Gly337Glu). This variant is present in population databases (rs767463971, gnomAD 0.008%). This variant has not been reported in the literature in individuals affected with BSCL2-related conditions. ClinVar contains an entry for this variant (Variation ID: 567989). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
