Truncations downstream of this position have been reported in association with McArdle disease in HGMD.The variant was absent in 251490 control chromosomes. c.2352C>A has been reported in the literature as a compound heterozygous genotype in and individual affected with Glycogen Storage Disease, Type V (also known as McArdle disease; Rubio_2007). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Four clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as pathogenic (n-=2), likely pathogenic (n=2). Based on the evidence outlined above, the variant was classified as likely pathogenic. - |
