Variant summary
Our verdict is Uncertain significance. The variant received 2 ACMG points: 7P and 5B. PM1PP2PP3_StrongBP6BS2
The NM_001370259.2(MEN1):c.710C>T(p.Ala237Val) variant causes a missense change involving the alteration of a conserved nucleotide. The variant allele was found at a frequency of 0.00000684 in 1,461,858 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Conflicting classifications of pathogenicity (no stars). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. A237T) has been classified as Uncertain significance.

The p.A237V variant (also known as c.710C>T), located in coding exon 3 of the MEN1 gene, results from a C to T substitution at nucleotide position 710. The alanine at codon 237 is replaced by valine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
