Members of this gene family bind folic acid and its reduced derivatives, and transport 5-methyltetrahydrofolate into cells. This gene product is a secreted protein that either anchors to membranes via a glycosyl-phosphatidylinositol linkage or exists in a soluble form. Mutations in this gene have been associated with neurodegeneration due to cerebral folate transport deficiency. Due to the presence of two promoters, multiple transcription start sites, and alternative splicing, multiple transcript variants encoding the same protein have been found for this gene. For these reasons, this variant has been classified as Pathogenic. Studies have shown that this premature translational stop signal alters FOLR1 gene expression (PMID: 22586289). ClinVar contains an entry for this variant (Variation ID: 16256). This premature translational stop signal has been observed in individual(s) with cerebral folate deficiency (PMID: 19732866). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This sequence change creates a premature translational stop signal (p.Cys175*) in the FOLR1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 83 amino acid(s) of the FOLR1 protein. - |
