This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 1579 of the MYO7A protein (p.Glu1579Lys). This variant is present in population databases (rs374766654, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with MYO7A-related conditions. ClinVar contains an entry for this variant (Variation ID: 504648). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt MYO7A protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.Glu1579Lys variant in MYO7A has not been previously reported in individual s with hearing loss or Usher syndrome, but has been identified in 8/ 126284 of E uropean chromosomes and 5/ 34374 of Latino chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP rs374766654). Althoug h this variant has been seen in the general population, its frequency is not hig h enough to rule out a pathogenic role.
