The p.M675V variant (also known as c.2023A>G), located in coding exon 18 of the MRE11A gene, results from an A to G substitution at nucleotide position 2023. The methionine at codon 675 is replaced by valine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces methionine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 675 of the MRE11 protein (p.Met675Val). This variant is present in population databases (rs756418071, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with MRE11-related conditions. ClinVar contains an entry for this variant (Variation ID: 820536).
