The p.R520H variant (also known as c.1559G>A), located in coding exon 13 of the MRE11A gene, results from a G to A substitution at nucleotide position 1559. The arginine at codon 520 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -

This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 520 of the MRE11 protein (p.Arg520His). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database.
