MRE11 (HGNC:7230): (MRE11 homolog, double strand break repair nuclease) This gene encodes a nuclear protein involved in homologous recombination, telomere length maintenance, and DNA double-strand break repair. By itself, the protein has 3' to 5' exonuclease activity and endonuclease activity. The protein forms a complex with the RAD50 homolog; this complex is required for nonhomologous joining of DNA ends and possesses increased single-stranded DNA endonuclease and 3' to 5' exonuclease activities. In conjunction with a DNA ligase, this protein promotes the joining of noncomplementary ends in vitro using short homologies near the ends of the DNA fragments. This gene has a pseudogene on chromosome 3. Alternative splicing of this gene results in two transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]

Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 250976 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.497C>T has been reported in the literature in at-least one individual affected with Cerebellar ataxia (example: Fogel_2014, Lee_2014). These report(s) do not provide unequivocal conclusions about association of the variant with Ataxia Telangiectasia-Like Disorder. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The proline at codon 166 is replaced by leucine, an amino acid with similar properties. This alteration was observed in conjunction with MRE11A c.168G>T in an individual with early onset ataxia, oculomotor apraxia, and extrapyradimal features and an affected brother; however, the phase (whether in cis or trans) of these two alterations was not specified (Fogel BL et al. JAMA Neurol. 2014 Oct;71:1237-46). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
