Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 250962 control chromosomes. c.856G>A has been reported in the literature in multiple individuals affected with classical PKU, mild PKU and MHP (eg. Liang_2014, Wang_2018, etc). These data indicate that the variant is very likely to be associated with disease. PAH activity from COS-1 cells transfected with the showed the variant to have <10% PAH activity compared to wild-type (Liang_2014). One diagnostic clinical lab and one expert panel have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. - |
