T-box genes encode transcription factors involved in the regulation of developmental processes. This gene is closely linked to related family member T-box 3 (ulnar mammary syndrome) on human chromosome 12. The encoded protein may play a role in heart development and specification of limb identity. Mutations in this gene have been associated with Holt-Oram syndrome, a developmental disorder affecting the heart and upper limbs. The serine at codon 261 is replaced by cysteine, an amino acid with dissimilar properties. This variant was previously reported in a family with features of Holt-Oram syndrome, and one study proposed this alteration may impact gene interactions involved in transcription and cardiac development (Brassington AM et al. Am J Hum Genet. 2003;73:74-85; Waldron L et al. Dev Cell. 2016;36(3):262-75). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
