The p.Q2283* variant (also known as c.6847C>T), located in coding exon 49 of the POLE gene, results from a C to T substitution at nucleotide position 6847. This changes the amino acid from a glutamine to a stop codon within coding exon 49. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.004% (greater than 25000 alleles tested) in our clinical cohort. This truncation occurs near the 3' terminus of POLE and impacts only the last 4 amino acids of the protein. The exact functional impact of these altered amino acids is unknown at this time. Since supporting evidence is limited at this time, the clinical significance of p.Q2283* remains unclear. -
