This sequence change replaces aspartic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 2115 of the POLE protein (p.Asp2115Gly). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with personal or family history of breast or ovarian cancer (PMID: 36427680). ClinVar contains an entry for this variant (Variation ID: 473796). The p.D2115G variant (also known as c.6344A>G), located in coding exon 46 of the POLE gene, results from an A to G substitution at nucleotide position 6344. The aspartic acid at codon 2115 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
