The p.H1410P variant (also known as c.4229A>C), located in coding exon 33 of the POLE gene, results from an A to C substitution at nucleotide position 4229. The histidine at codon 1410 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
