The p.L1110I variant (also known as c.3328C>A), located in coding exon 27 of the POLE gene, results from a C to A substitution at nucleotide position 3328. The leucine at codon 1110 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
