The aspartic acid at codon 301 is replaced by glycine, an amino acid with similar properties. This alteration was identified in a caucasian male with MSI-L colon cancer diagnosed at age 58 and no reported history of polyps from a cohort of 1046 individuals with familal colorectal cancer (Raskin L et al. Oncotarget. 2017 Nov;8:93450-93463). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be inconclusive by in silico analyses. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
