The lysine at codon 284 is replaced by glutamic acid, an amino acid with similar properties. In a study of 51 individuals with multiple colonic adenomas from 48 families who underwent whole-exome sequencing, this variant was identified in a patient with a mix of 30 adenomatous, hyperplastic, and serrated polyps who did not have first degree relatives with colorectal cancer or polyps (Weren RD et al. Nat. Genet. 2015 Jun;47:668-71). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
