The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.850A>G has been reported in the literature in settings of whole exome sequencing (WES) performed in at-least one individual affected with Colorectal Cancer highly suspected of a hereditary cause (example, Weren_2015). These report(s) do not provide unequivocal conclusions about association of the variant with Colorectal Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. In a study of 51 individuals with multiple colonic adenomas from 48 families who underwent whole-exome sequencing, this variant was identified in a patient with a mix of 30 adenomatous, hyperplastic, and serrated polyps who did not have first degree relatives with colorectal cancer or polyps (Weren RD et al. Nat. Genet. 2015 Jun;47:668-71). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
