This sequence change creates a premature translational stop signal (p.Pro269Leufs*26) in the POLE gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in POLE are known to be pathogenic (PMID: 23230001, 25948378, 30503519). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with endometrial cancer (PMID: 33001133). ClinVar contains an entry for this variant (Variation ID: 473833). For these reasons, this variant has been classified as Pathogenic. -

Frameshift variant predicted to result in protein truncation or nonsense mediated decay in a gene for which loss of function is a known mechanism of disease; Not observed at significant frequency in large population cohorts (gnomAD); Identified in an individual with endometrial cancer (PMID: 33001133); This variant is associated with the following publications: (PMID: 33001133) -

The c.806delC variant, located in coding exon 9 of the POLE gene, results from a deletion of one nucleotide at nucleotide position 806, causing a translational frameshift with a predicted alternate stop codon (p.P269Lfs*26). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. Loss-of-function variants subject to nonsense mediated decay (NMD) in POLE are known to cause POLE deficiency; however, such associations with POLE-related polymerase proofreading-associated polyposis (PPAP) have not been reported. Based on the supporting evidence, this alteration is pathogenic for POLE deficiency; however, the association of this alteration with POLE-related PPAP is unknown. -
